بررسی اثر غلظت فنل بر رشد، تنفس و تشکیل بیوفیلم باکتری‌های تجزیه کننده فنل در پساب کارخانه ذوب‌آهن اصفهان

چکیده

ترکیبات فنلی، فنل و مشتقات فنل، آلوده کننده‌های محیطی هستند که در پساب‌های صنعتی مختلف مانند تبدیلات زغال سنگ، پالایشگاه نفت، کارخانه‌های شیمیایی و پتروشیمی وجود دارند. حضور این ترکیبات در پساب  کارخانه ذوب‌آهن اصفهان خطر جدی برای محیط زیست منطقه در پی دارد. بهترین و کم هزینه‌ترین روش تصفیه پساب‌های آلوده به فنل و ترکیبات فنلی استفاده  از روش‌های تصفیه زیستی است. در این تحقیق، 15 سویه باکتریایی تجزیه کننده فنل از مکان‌های مختلف کارخانه ذوب‌آهن اصفهان جدا سازی گردید . از بین این 15 سویه باکتریایی، 5 سویه به عنوان سویه‌های غالب با نام سویه‌های  B3, C1, C4, C7, D14 شناخته شدند، که قابلیت بالایی برای حذف فنل نشان می‌دادند. سپس اثر غلظت‌های مختلف فنل (900-100 میلی گرم بر لیتر) بر میزان رشد، تنفس و تشکیل بیو فیلم این 5 سویه بررسی شد . نتایج حاکی از آن بود که کلیه سویه‌ها تا غلظت 500 میلی‌گرم  بر لیتر فنل، رشد و حذف فنل مناسبی نشان می‌دادند؛ اما در این بین میزان رشد سویه B3 در غلظت 300 میلی‌گرم بر لیتر بالاتر از چهار سویه دیگر بود و همین سویه، بالاترین بیو فیلم را در این غلظت تشکیل می‌داد. تنفس سویه‌ها در غلظت‌های فزاینده فنل متناسب با رشد آن‌هاست به طوری که اکثر سویه‌ها بالاترین تنفس را در غلظت 500-400  میلی‌گرم بر لیتر فنل نشان می‌دادند و در همین غلظت بالاترین رشد و بیو فیلم را داشتند. دوره انکوباسیون نیز تأثیر به سزایی بر حذف فنل سویه‌ها داشت، بدین صورت که در دوره انکوباسیون 24 ساعته تنها تا غلظت 500 میلی‌گرم بر لیتر تجزیه می‌گردید؛ اما با افزایش این زمان به 48 ساعت، فنل به طور کامل تا غلظت 900 میلی‌گرم بر لیتر تجزیه می‌گردید. با به کارگیری این سویه‌ها در حوضچه‌های تصفیه فنل کارخانه ذوب‌آهن می‌توان میزان آلودگی فنلی این کارخانه را به طور قابل توجهی کاهش داد.

Phenol and phenolic compounds are environmental pollutants that are found in industrial wastewater of oil refineries, coal tar, and chemical plants. Presences of these compounds in Isfahan steal plant have serious consequences for regional environment. Biodegradation is the best and economic method for refinement of phenol contaminated sites. In this research 15 phenol-degrading bacteria strains were isolated from different sites of Isfahan steal plant wastewater. Five dominant strains named as B3, C1, C4, C7 and D14 that had high capacity to eliminate phenol. Effects of different concentrations of phenol (100-900 mg/ml) on growth. Respiration and biofilm formation of 5 dominant strains were investigated. All strains grown well up to 500 mg/l of phenol concentration and eliminated it. Strains B3 had the most growth and biofilm formation on 300 mg/l.  The majority strains had the best respiration growth and biofilm formation in 400-500 mg/l of phenol concentration. At 24 hours incubation time up to 500 mg/l phenol of phenol was degraded and at 48 hours incubation time 900 mg/l of phenol was degraded. By using these strains it could be possible to reduce phenol pollutant in phenol refinement pools of the Isfahan steal plant.
