شناسایی و اندازه گیری مقادیر کلروفیل هایa و b در سدهای کرج، طالقان و لتیان با استفاده از روش کروماتوگرافی مایع با کارایی بالا ( HPLC/UV-VIS)

چکیده

در این تحقیق شناسایی و اندازه‌گیری مقدار کلروفیل‌هایa  و b با استفاده از روش کروماتوگرافی مایع با کارایی بالا (UV-VIS/HPLC) انجام شد. منحنی‌های کالیبراسیون برای کلروفیل‌های a و b از نوع خطی و با ضریب رگرسیون (999/0 < r ) به‌دست آمد. نمونه‌برداری از سدهای کرج، طالقان و لتیان از آذرماه 1390 تا آذرماه 1391 به‌صورت فصلی انجام شد. میانگین مقدار کلروفیل a و b طی چهار فصل سال در سد کرج به‌ترتیب ppb5/4 و  ppb028/0 ، طالقانppb  4/2 و ppb04/0 و لتیان  ppb3 وppb 03/0 به‌دست آمد. بنابراین روش کروماتوگرافی مایع با کارایی بالا یک روش معتبر و مطمئن برای اندازه‌گیری نمونه‌های دارای کلروفیل در حد  ppbاست.

In this study detection and measurment of chlorophylls a and b by using high performance liquid chromatography (HPLC/UV-VIS) in  Karaj,Taleghan and Latian Reservoirs were performed.The calibration graphs were linear with very good correlation coefficients (r > 0.999) for chlorophylls a and b. Samples were collected from Karaj, Taleghan, and Latian Reservoirs in duplicate for  years 2012-2013. The avarage chlorophyll a and b in Karaj Resevoir were 4.5 and 0.028 µg/l, Taleghan Reservoir 2.4 and 0.04 µg/l, and Latian Reservoir 3 and 0.03 µg/l ,respectively, and in some cases less than detection limit (LOD). This study showed that high performance liquid chromatography is a valid method for determination of chlorophylls a and b in natural water samples in microgram per liter.
