اثر بعضی پارامترهای فیزیکی و شیمیایی بر رشد باکتری آئروموناس و باکتری‌های هتروتروف (HPC) در شبکه توزیع آب آشامیدنی اصفهان

چکیده

آئروموناس از جمله باکتری‌های بی‌هوازی اختیاری، گرم منفی، بدون اسپور و میله‌ای شکل و فرصت طلب است که می‌تواند باعث ایجاد عفونتهای سیستمیک، و زخم و اسهال در انسان شود. جمعیت نسبتاً بالای میکروبی در شبکه‌های توزیع آب آشامیدنی نه تنها از لحاظ تأثیرگذاری بر سلامت مصرف کننده مورد توجه است، بلکه تشخیص باکتری شاخص کلیفرم را نیز با خطا مواجه می‌کند. در این تحقیق ارتباط رشد باکتری‌های هتروتروف و آئروموناس با پارامترهای pH، دما، کدورت، کلر آزاد باقیمانده و اکسیژن محلول سنجیده شد. به منظور شمارش باکتری‌های آئروموناس، برای اولین بار در کشور طبق دستورالعمل 1605 EPA (2001) از محیط کشتADA-V در مرحله احتمالی و پس از قرار دادن در انکوباتور به مدت یک شب با محیط کشت نوترینت آگار از تست‌های اکسیداز، تخمیر تری‌هالوز و تست ایندول در مرحله تأییدی استفاده شد. برای شمارش باکتری‌های HPC ، از محیط R2A استفاده شد و بقیه فاکتورها نیز طبق روش استاندارد اندازه‌گیری شدند. نتایج نشان داد که در دما و کدورت بالاتر و pH پایین‌تر میزان موارد مثبت باکتری‌های آئروموناس و HPC افزایش می‌یابد و با افزایش کلر آزاد باقیمانده و اکسیژن محلول موارد مثبت باکتری‌های آئروموناس و HPC کاهش می‌یابد و در مقادیر بالاتر از mg/L 0/2 کلر باقیمانده آزاد هیچ گونه مورد مثبتی از آئروموناس مشاهده نشد.

Aeromonas is one the gram – negative , non spor – formating rod shaping , facultatively anaerobic and opportunistic bactria that can cause systematic infections, leision and diarrhoea in human. Fairly high bactrial population in distribution system is not only of concern because of affecting consumer health but also it makes it difficult to enumerate coliform bacterium indicator. So, the relationships between aeromonas and heterotrophic bacteria growth with pH, temperature, turbidity, free residual cholornie and DO were determined in this study. ADA- V media was used in presumptive stage to count aeromonas bacteria for the first time in Iran on the basis of 1605 EPA (2001) method and used oxidase tests, trehalose fermentation and indol test in confirmative stage. R2A media was used to count HPC bacteria and other factors measured on the basis of standards. The results showed that positive cases of aeromonas bacteria and HPC increase in higher temperature and turbidity and lower pH. In contrast, positive cases of aeromonas bacteria and HPC decrease while free residual cholorine and DO increase. In addition,no positive case of aeromonas was observed in more than 0.2 mg/L concentration of free residual cholorine.
