بررسی تجزیه‌زیستی پارانیتروفنل در کشت لرزان و بیوراکتور هواگرد خارجی توسط باکتری آلکالیجنس فایکالیس

چکیده

در این مطالعه برای اولین بار توانایی تجزیه زیستی پارانیتروفنل توسط باکتری آلکالیجنس فایکالیس در یک پساب نمونه در بیوراکتور هواگرد بررسی شد. در ابتدا اثر عواملی چون منبع نیتروژنی (عصاره مخمر، پپتون و اوره)، قلیاییت محیط (pH برابر 6، 7 و 8) و غلظت اولیه نیتروفنل (50، 75، 100 و 125 میلی‌گرم در لیتر) بر رشد باکتری و حذف نیتروفنل در کشت لرزان بررسی گردید. سپس عملکرد بیوراکتور هواگرد در حذف نیتروفنل تحت شرایط مناسب به‌دست آمده در کشت لرزان در غلظتهای اولیه 50، 75 و 100 میلی‌گرم در لیتر و نرخهای هوادهی متفاوت 1، 2 و 4 لیتر در دقیقه مورد مطالعه قرار گرفت. نتایج کشت لرزان بیانگر آن است که استفاده از عصاره مخمر و pH 8 منجر به بالاترین مقدار توده‌ زیستی و میزان حذف نیتروفنل می‌گردند. نتایج بیوراکتور هواگرد در تمامی نرخهای هوادهی، نشان دهنده درصد حذف بیشتر نیتروفنل و کاهش زمان حذف نسبت به کشت لرزان است. بیشترین میزان حذف در کشت لرزان و بیوراکتور هواگرد در غلظت50 میلی‌گرم در لیتر به‌ترتیب به‌میزان 40 و60 درصد به‌دست آمد. در میان نرخهای هوادهی، 2 لیتر در دقیقه، بیشترین میزان حذف را در همه غلظتهای نیتروفنل در بیوراکتور نشان می‌دهد.

In this study, removal of para-nitrophenol (PNP) from synthetic wastewater by Alcaligens faecalis was investigated for the first time in an external airlift bioreactor. The effect of factors such as pH (6, 7 and 8) and nitrogen source (yeast extract, peptone and urea) as well as initial PNP concentration (50, 75, 100 and 125 mg/lit) on PNP removal and bacterial growth was first examined in shake flask experiments. Then, performance of airlift bioreactor was studied at various aeration rates (1, 2 and 4 l/min) and initial PNP concentrations (50, 75 and 100 mg/lit). In shake flask experiments, yeast extract and pH value of 8 led to the highest bacterial growth and PNP removal. In all bioreactor experiments, PNP removal was higher and biodegradation time was lower compared to shake flask experiments. The maximum of 40 and 60% PNP removal were obtained in shake flask and bioreactor experiments at 50 mg/l initial PNP concentration respectively. Among examined aeration rates, 2 L/min resulted in the highest PNP removal percentage at all initial PNP concentration.
