جداسازی و شناسایی باکتری‌های تجزیه‌کننده فنل از پساب کارخانه گل‌گهر سیرجان

چکیده

فنل و مشتقات آن ترکیباتی به‌شدت سمی می‌باشند که به‌راحتی می‌توان آنها را از پساب‌های صنایع مختلفی مانند پالایشگاه‌های نفت، صنایع پتروشیمی، معادن به‌ویژه زغال سنگ و کارخانه‌های مواد شیمیایی حذف کرد. ورود این مواد به محیط‌ زیست باعث آلودگی‌های شدید به‌ویژه در منابع آبی می‌شود. در گذشته از روش‌های فیزیکوشیمیایی برای حذف فنل و مشتقات آن استفاده می‌شد اما امروزه تصفیه زیستی در اولویت قرار دارد. هدف از این پژوهش شناسایی و جداسازی باکتری‌های تجزیه‌کننده فنل از پساب کارخانه گل‌گهر سیرجان بود. در این تحقیق برای جداسازی باکتری‌های تجزیه کننده فنل، نمونه‌برداری از مناطق مختلف پساب کارخانه گل‌گهر سیرجان صورت گرفت و با استفاده از روش‌های غنی‌سازی در محیط بوشنل- هاس همراه با فنل به‌عنوان منبع کربن، باکتری‌های تجزیه کننده فنل جداسازی شدند. باکتری‌های برتر در تجزیه فنل با استفاده از پرایمرهای ویژه ژن S rRNA16 و انجام واکنش زنجیره‌ای چندگانه و در نهایت تعیین توالی باند حاصله شناسایی شدند. در طی این تحقیق، ۱۷ سویه باکتریایی تجزیه‌کننده‌ فنل از نمونه‌های خاک و پساب جمع‌آوری شده از مناطق مختلف معدن جداسازی شد. با انجام آزمون‌های غربالگری، تعداد چهار سویه به‌عنوان سویه‌های برتر انتخاب شدند. بالاترین غلظتی از فنل که به این باکتری‌ها تجزیه می‌شد، غلظت ۴/۰ گرم در لیتر بود. نتایج حاصل از شناسایی مولکولی نشان داد که این باکتری‌های برتر تجزیه‌کننده فنل متعلق به جنس‌های سودوموناس اس پی و نیتراتیرداکتر اس پی و سالجنتی باکتر اس پی هستند. در مجموع نتایج این تحقیق نشان داد پساب کارخانه گل‌گهر سیرجان دارای تعداد زیادی از باکتری‌های تجزیه کننده فنل به‌ویژه جنس‌های مذکور است و با به‌کارگیری این باکتری‌ها در پساب حاصل از کارخانه گل‌گهر می‌توان میزان آلودگی این کارخانه را به‌طور قابل توجهی کاهش داد.

Phenol and phenolic compounds are highly toxic substances that are found as monoaromatic compounds in various industrial effluents from oil refineries, petrochemical plants, (coal) mines, and phenol resin plants. Their discharge into the environment, especially in water resources, causes serious toxicity. Traditionally, physicochemical methods have been used for the removal of phenol and phenolic compounds. Nowadays, bioremediation is known to be the best method for phenol removal from wastewater. The objective of the present study was twofold: isolation and identification of phenol degrading bacteria in the effluent from Golgohar Mine in Sirjan. For this purpose, samples were collected from different sections at Golgohar Mine and its effluent. Phenol degrading bacteria were isolated via enrichment of the samples in the Bushnell Hass medium with phenol used as the only source of carbon and energy. The predominant phenol degrader bacteria were selected by measuring turbidity at 600 nm. The bacteria were subsequently identified by amplification of 16S rRNA with specific primers and PCR sequencing. In this study, 17 strains of phenol degrader bacteria were isolated in soil and wastewater samples collected from different zones of the mine. Screening methods confirmed that 4 strains exhibit a better capability for phenol degradation as evidenced by their capability to degrade 0.4 g/l of phenol. Molecular identification showed that these bacteria belong to the species Pesudomonas sp, Nitrratireductor sp., and Salegentibacter sp. The results also show that the effluent from Golgohar Mine in Sirjan contains many phenol degrading bacteria. The use of these bacteria in the treatment process may lead to a significant reduction in phenol pollution in the mineral effluent.
